He was born on December 24, 1940, in Brooklyn, New York to Stephen A Fauci and Eugenia Lillian. His dad studied pharmacy at Columbia University and later owned his own practice. The whole family worked at the pharmacy, with Fauci delivering prescriptions and working the register.

Fauci attended Regis High School, a prestigious all-boys Jesuit high school in Manhattan. He went on to earn his undergraduate degree from the College of the Holy Cross in Worcester, Massachusetts and his medical degree from Cornell University Medical College.
